The film I remember laughing loudest at in a theater in my life was There's Something About Mary. The American football subplot may be slightly off if you aren't familiar with it but it is genuinely unbelievably hilarious -- LOL funny beginning to end.

The first 30 minutes of the South Park film is also really, really LOL good as is a lot of Team America: World Police.

Reading through this there are quite a few films I found funny at first viewing but wouldn't if I watched them again. I think with comedy and horror, once you've seen them once the shock/surprise factor is gone. I would suggest the python films, but they've been ruined for me by unfunny cunts reciting the scripts from memory.
