Madame Tussauds is expensive and probably a bit naff.
Tower of London definitely, and the river ferry is a decent idea. Go down to Westminster and then walk round to Buckingham Palace, it's not far, and you can go past the Abbey or up Whitehall/Trafalgar Square/The Mall if you want. St James' Park (next to the Palace) is my favourite park in the centre.
EB's comments about the London Bridge area are spot on, and it's not far along to the Globe, Tate Modern and the Golden Hind replica.
Harrods is a bit further away, but doable. Never been there!
When it's raining, the British Museum may be a decent fallback to see what we've stolen, sorry, acquired from other places.
if it's fine of a evening, and you don't mind standing, the Globe will have evening performances for about £5 (may be a bit worthy for the kids!)
