There was a similar thread a few weeks ago when a lad from Canada was about to visit Manchester. We ended up meeting up with him, and actually ended up learning a few things through visiting places through a tourist's eyes.
Without listing every museum, pub & eatery which others have mentioned, here's some of the places we took our Canadian friend.
Pubs: Cask (x 2), Crown & Kettle, Seven Brothers, The Old Wellington (also for a meal), Piccadilly Tap & Northern Monk.
One of the things he mentioned was that he'd never experienced pubs which only sold drinks before, with all the establishments back home being required by law to also sell food.
Food: Bundobust (fantastic array of Indian Street food).
Places of interest: As well as the many museums others have mentioned, it's worth having a look around Castlefield (the marina, Roman ruins & Castlefield Viaduct National Trust site) & "old Manchester" (Cheetham's, the Cathedral, Shambles Square).
